ทุกอย่างในโรงแรมทรานซิลเวเนียดูจะเปลี่ยนแปลงไปในทิศทางที่ดีขึ้น กฎเหล็กของท่าน เคานต์ แดรกคิวลา (อดัม แซนด์เลอร์) เริ่มผ่อนผัน จากโรงแรมที่ต้อนรับเฉพาะแขกที่เป็นภูตผีปีศาจ ตอนนี้ก็ยอมเปิดประตูให้แขกมนุษย์มาพักอาศัยแล้ว แม้ทุกอย่างจะดูดีไปหมด แต่ท่านเคานต์ยังมีเรื่องให้กลุ่มใจอยู่หลังฝาโลง เพราะ เดนนิส (แอชเชอร์ บลิงก์ออฟ) หลานชายครึ่งผีดูดเลือดครึ่งมนุษย์กลับไม่มีทีท่าว่าเป็นผีดูดเลือดเลยสักนิด

ตอนที่ มาวิส (เซเลนา โกเมซ) ลูกสาวผีดูดเลือดของท่านเคานต์ไม่อยู่ ท่านเคานต์จึงรวบรวมพรรคพวก ทั้ง แฟรงก์ หรือ แฟรงเกนสไตน์ (เควิน เจมส์) มนุษย์หมาป่า เวย์น (สตีฟ บุสเชมี) มัมมี เมอร์เรย์ (คีแกน-ไมเคิล คีย์) และมนุษย์ล่องหน กริฟฟิน (เดวิด สเปด) ให้ช่วยกันจัดค่ายฝึกสำหรับภูติผีฝึกหัดแก่เดนนิส

เหล่าพี่เลี้ยงค่ายไม่รู้เลยว่า วลาด (เมล บรูกส์) คุณพ่อที่แก่แสนแก่และสุดจะหัวโบราณของท่านเคานต์กำลังจะเดินทางมาโรงแรมสุดหรู ตอนที่วลาดรู้ว่าเหลนของตัวเองเป็นเลือดผสม แถมต้องมารับรู้ว่าโรงแรมได้ให้การต้อนรับอย่างอบอุ่นแก่พวกมนุษย์ด้วยแล้ว เรื่องราวจึงยิ่งวุ่นหนักเข้าไปอีก

Everything seems to be changing for the better at Hotel Transylvania... Dracula's rigid monster-only hotel policy has finally relaxed, opening up its doors to human guests. But behind closed coffins, Drac is worried that his adorable half-human, half-vampire grandson, Dennis, isn't showing signs of being a vampire.

So while Mavis is busy visiting her human in-laws with Johnny - and in for a major cultural shock of her own - "Vampa" Drac enlists his friends Frank, Murray, Wayne and Griffin to put Dennis through a "monster-in-training" boot camp.

But little do they know that Drac's grumpy and very old, old, old school dad Vlad is about to pay a family visit to the hotel. And when Vlad finds out that his great-grandson is not a pure blood - and humans are now welcome at Hotel Transylvania - things are going to get batty!
